چکیده
Table 1. Numeric Values of Input Quantities of the Model for the Reference State The data are grouped into concentrations of pathway components, dissociation constants of protein complexes, concentration ratios, fl uxes and fl ux ratios, and characteristic times of selected processes. Experimental evidence for these data is discussed in the text. From these data, the following rates and rate constants are calculated: ν12 = 0.42 nM • min (rate of β-catenin synthesis), ν14 = 8.2 • 10 • nM • min (rate of axin synthesis), k4 = 0.27 min, k5 = 0.13 min, k6 = 9.1 • 10 nM • min, k–6 = 0.91 • nM • min, k9 = 210 min, k10 = 210 min, k11 = 0.42 min, k13 = 2.6 • 10 –4 min, k15 = 0.17 • min. The Roles of APC and Axin Derived from Experimental and Theoretical Analysis of the Wnt Pathway
Wnt signaling plays an important role in both oncogenesis and development. Activation of the Wnt pathway results in stabilization of the transcriptional coactivator beta-catenin. Recent studies have demonstrated that axin, which coordinates beta-catenin degradation, is itself degraded. Although the key molecules required for transducing a Wnt signal have been identified, a quantitative understa...

Background and Aim: Different regulation processes have an effect on osteoblastic differentiation of mesenchymal stem cells (MSCs), and among them Wnt signaling pathway is particularly desirable. In Wnt signaling pathway, Adenomatous Polyposis Coli (APC) bind to β-catenin and induce its degradation, thereby acting as a negative regulator of canonical Wnt pathway. In this study, gene expression ...
متن کاملThe roles of intrinsic disorder in orchestrating the Wnt-pathway.
The canonical Wnt-pathway plays a number of crucial roles in the development of organism. Malfunctions of this pathway lead to various diseases including cancer. In the inactivated state, this pathway involves five proteins, Axin, CKI-α, GSK-3β, APC, and β-catenin. We analyzed these proteins by a number of computational tools, such as PONDR(r)VLXT, PONDR(r)VSL2, MoRF-II predictor and Hydrophobi...
متن کاملAxin phosphorylation in both Wnt-off and Wnt-on states requires the tumor suppressor APC
The aberrant activation of Wnt signal transduction initiates the development of 90% of colorectal cancers, the majority of which arise from inactivation of the tumor suppressor Adenomatous polyposis coli (APC). In the classical model for Wnt signaling, the primary role of APC is to act, together with the concentration-limiting scaffold protein Axin, in a "destruction complex" that directs the p...
متن کاملWnt/Wingless Pathway Activation Is Promoted by a Critical Threshold of Axin Maintained by the Tumor Suppressor APC and the ADP-Ribose Polymerase Tankyrase.
Wnt/β-catenin signal transduction directs metazoan development and is deregulated in numerous human congenital disorders and cancers. In the absence of Wnt stimulation, a multiprotein "destruction complex," assembled by the scaffold protein Axin, targets the key transcriptional activator β-catenin for proteolysis. Axin is maintained at very low levels that limit destruction complex activity, a ...
